Melanin-sanshool refers to a bioinspired nanoparticle formulation in which sanshool, a naturally occurring molecule from Zanthoxylum species known for its antioxidant and photoprotective properties, is combined with synthetic or natural melanin-like materials. This combination is achieved via boron esterification interactions to form stable nanoparticles (melanin–S NPs). The resulting formulation exhibits improved stability and enhanced photoprotective efficiency compared to sanshool alone. Melanin provides broad-spectrum ultraviolet absorption and antioxidative capacity, while sanshool contributes additional antioxidant activity. The primary mechanism of action involves the synergistic enhancement of UV protection and reduction of oxidative stress in skin models. This approach aims to overcome the inherent instability and potential skin permeation risks associated with free sanshool by leveraging the protective matrix provided by melanin-like polymers[1][2]. The main application under investigation is as a topical agent for preventing photodamage (e.g., sun-induced skin damage), with demonstrated efficacy in vitro and in animal models[1].
