Overview

Melanoma cell lysate-pulsed autologous dendritic cell vaccine is an investigational autologous cellular immunotherapy designed to treat advanced malignancies, particularly metastatic melanoma and MAGE-A-expressing colorectal cancer. The therapeutic process involves harvesting a patient's peripheral blood mononuclear cells (PBMCs) and differentiating them ex vivo into dendritic cells (DCs) using growth factors such as GM-CSF and IL-4. These DCs are then pulsed with an allogeneic melanoma cell lysate (such as the DDM-1 line) which is rich in cancer/testis antigens, specifically the MAGE-A family (A1, A3, A4, A6, A10, and A12). To ensure high immunogenicity, the DCs are matured using a cytokine cocktail typically comprising IL-1β, TNF-α, IL-6, and prostaglandin E2 (PGE2), resulting in a phenotype with high expression of CD83 and CCR7 for optimized lymph node trafficking. Once administered intradermally, these mature DCs present tumor-associated antigens to the patient's T cells, aiming to induce a robust cytotoxic T-lymphocyte response and downregulate immunosuppressive regulatory T cells (Tregs) within the tumor microenvironment.
