The melanoma helper peptide vaccine is a multivalent therapeutic cancer vaccine composed of six synthetic peptides derived from melanoma-associated antigens (including cancer-testis antigens and melanocytic differentiation proteins) designed to stimulate CD4+ T cell (helper T cell) responses against melanoma. The formulation may also include an adjuvant such as incomplete Freund’s adjuvant or a tetanus toxoid-derived peptide to enhance immunogenicity. The primary mechanism is the induction of robust class II MHC-restricted immune responses that support anti-tumor activity by promoting both direct and indirect activation of cytotoxic T cells and antibody production. This approach has been shown in clinical trials to be safe, immunogenic in most patients with advanced or high-risk resected melanoma, and associated with improved overall survival when combined with other therapies such as PD-1 blockade[1][3][5][6].
