Memit is a novel small molecule prodrug of memantine designed for the treatment of Alzheimer's disease. It is a hybrid molecule synthesized by replacing the free amine group of memantine with an isothiocyanate functionality, which serves as a hydrogen sulfide (H2S) donor. Upon administration, memit releases H2S through a cysteine-mediated mechanism, simultaneously regenerating the parent drug, memantine. This dual-action approach aims to combine the NMDA receptor antagonism of memantine with the potent anti-inflammatory, antioxidant, and neuroprotective properties of H2S. Preclinical in vitro studies have demonstrated that memit reduces reactive oxygen species (ROS) production, inhibits Aβ(1-42) self-induced aggregation, protects against Aβ oligomer-induced neuronal damage, and promotes autophagy, addressing multiple pathological pathways associated with neurodegeneration.
