Overview

MEN1112 is a humanized, Fc-engineered, de-fucosylated monoclonal immunoglobulin G1 (IgG1) antibody that targets the bone marrow stromal cell antigen 1 (BST1/CD157). It was developed as an antineoplastic agent with the primary mechanism of action being antibody-dependent cellular cytotoxicity (ADCC), particularly through natural killer (NK) cell-mediated cytotoxicity. The drug was designed to enhance immune-mediated killing of CD157-expressing cells and has shown potent in vitro and ex vivo activity against acute myeloid leukemia (AML) cells. Developed by Menarini and Oxford BioTherapeutics, it reached phase I clinical trials for relapsed or refractory AML but development for this indication has been discontinued[1][3][5][6][8].
