Menatetrenone disodium diphosphate is a water-soluble, injectable small molecule analogue of vitamin K2 (menatetrenone, also known as MK-4). Developed by Guangdong Aimin Pharmaceutical Co., Ltd., it serves as a cofactor for the enzyme gamma-glutamyl carboxylase, which is essential for the post-translational gamma-carboxylation of glutamate residues in vitamin K-dependent proteins. This process is critical for the activation of coagulation factors II (prothrombin), VII, IX, and X, making the drug a potential therapeutic for hypoprothrombinemia and biliary colic. The disodium diphosphate salt formulation allows for parenteral administration, overcoming the solubility limitations of natural fat-soluble vitamin K2. It is currently in Phase 1 clinical development in China, with studies focused on characterizing its pharmacokinetic profile, safety, and dose linearity in healthy adult volunteers.
