The Chiron MenACWY Ad-Conjugate Vaccine is an investigational quadrivalent meningococcal conjugate vaccine developed by Chiron (later acquired by Novartis, and subsequently GSK). It is designed to provide active immunization against invasive meningococcal disease caused by *Neisseria meningitidis* serogroups A, C, W-135, and Y. The vaccine consists of the capsular polysaccharides of these four serogroups, each individually conjugated to the CRM197 carrier protein, a non-toxic mutant of diphtheria toxin. The "Ad-Conjugate" designation specifically refers to the formulation containing the MF59 adjuvant, an oil-in-water emulsion intended to enhance the immune response, particularly in infants and young children. This adjuvanted version was evaluated in Phase II clinical trials, where it demonstrated robust serum bactericidal antibody (SBA) responses compared to polysaccharide vaccines. While the non-adjuvanted version of this conjugate vaccine was eventually approved and marketed as Menveo, the adjuvanted formulation remained an investigational candidate.
