Merimepodib is an orally bioavailable small molecule that acts as a noncompetitive inhibitor of inosine monophosphate dehydrogenase (IMPDH), the enzyme responsible for the rate-limiting step in de novo guanine nucleotide synthesis. By inhibiting IMPDH, merimepodib reduces intracellular guanosine triphosphate (GTP) levels required for DNA and RNA synthesis, resulting in antiproliferative and antiviral effects. It has demonstrated activity against hepatitis C virus (HCV) and a range of other viruses including Zika, Ebola, Lassa, Chikungunya, and Junin viruses. Merimepodib was initially developed by Vertex Pharmaceuticals primarily for HCV infection but has also been investigated for other viral diseases such as COVID‑19 and psoriasis[1][2][3][4][5][6][7].
