mERK2 long peptide is a neoantigen-based cancer vaccine candidate targeting a specific mutation in the Extracellular Signal-Regulated Kinase 2 (ERK2), also known as Mitogen-Activated Protein Kinase 1 (MAPK1). This peptide was identified as a tumor-specific antigen in the CMS5a fibrosarcoma cell line, which is characterized by its resistance to immune checkpoint inhibitor (ICI) therapy. In development by Asahi Kasei and Mie University, the peptide is typically encapsulated within cholesterol-conjugated hyaluronic acid (HA) nanogels. This delivery system is designed to enhance targeting to draining lymph nodes and facilitate uptake by antigen-presenting cells such as macrophages and dendritic cells. When co-administered with an adjuvant like CpG oligoDNA, the vaccine induces a potent antigen-specific CD8+ cytotoxic T lymphocyte (CTL) response, demonstrating anti-tumor activity in preclinical models of "cold" tumors.
