Overview

MES-Fudan University-triple-negative breast cancer refers to the therapeutic strategy and associated drug regimens for the Mesenchymal-like (MES) subtype of triple-negative breast cancer (TNBC) as defined in the FUTURE (Fudan University Triple-negative breast cancer Umbrella REsearch) trial (NCT03805399). Developed by Fudan University, this precision medicine approach stratifies MES-subtype patients based on PI3K/AKT pathway activation. Patients without PI3K/AKT activation are treated with a combination of famitinib (a multi-kinase inhibitor targeting VEGFR, PDGFR, and c-Kit) and etoposide (VP-16). Patients with PI3K/AKT activation receive an mTOR inhibitor (typically everolimus) in combination with nab-paclitaxel. The MES subtype is characterized by high expression of genes involved in the epithelial-mesenchymal transition and angiogenesis, which these regimens are designed to target.
