MES1022 is an investigational, bone-selective EP4 receptor agonist prodrug being developed by Mesentech for bone regeneration and musculoskeletal disorders. It is a drug-conjugate prodrug that selectively distributes to bone, where it slowly releases a potent prostaglandin E2‑mimetic that activates the prostaglandin E2 receptor EP4, driving anabolic bone formation and secondary muscle growth while limiting systemic exposure and off‑target effects.[1][3][4][7][10][12] MES1022 is being advanced as a subcutaneous, once‑weekly therapy to accelerate fracture healing and potentially treat metabolic bone diseases, brittle bone disease, and bone loss associated with Duchenne muscular dystrophy, with Phase 1 trials completed or ongoing in patients with bone fractures.[1][2][3][4][7][10][11][14]
