Mesenchymal stem cell + HA-CaSO4 + BMP-2 is an investigational combination regenerative therapy developed by Universitas Indonesia for the treatment of critical-sized bone defects (CSDs), which are bone gaps exceeding 2.5 cm that typically fail to heal spontaneously. The approach is based on the 'diamond concept' of bone healing, which integrates four essential components: osteogenic autologous bone marrow-derived mesenchymal stem cells (BM-MSCs), an osteoconductive hydroxyapatite-calcium sulfate (HA-CaSO4) scaffold, an osteoinductive growth factor (bone morphogenetic protein-2 or BMP-2), and mechanical stability provided by fixation implants. By combining these elements, the therapy aims to induce robust osteogenesis and bridge large defects, offering a potential alternative to complex surgical procedures like vascularized bone grafting or bone transport.
