Mesenchymal stem cells Hospital de Cruces is an investigational allogeneic cell therapy developed by Hospital de Cruces and the Biocruces Bizkaia Health Research Institute for the treatment of Osteogenesis Imperfecta (OI). The therapy consists of HLA-matched mesenchymal stem cells (MSCs) derived from healthy sibling donors. These cells are intended to act as progenitors for osteoblasts, capable of secreting functional type I collagen to address the underlying genetic defect caused by mutations in the COL1A1 or COL1A2 genes. Clinical findings from the Phase I TERCELOI trial suggest that the therapy primarily functions through a pro-osteogenic paracrine mechanism rather than direct cell engraftment, resulting in improved bone formation, mineralization, and a reduction in fracture frequency in pediatric patients.
