MESO CAR-NK cells are genetically engineered natural killer (NK) cells that express a chimeric antigen receptor (CAR) targeting mesothelin, a tumor-associated antigen overexpressed in several cancers such as cervical, pancreatic, and ovarian cancer. The therapy involves introducing a third-generation anti-mesothelin CAR into NK-92 cell lines or primary human NK cells using alpharetroviral SIN vectors. These modified NK cells demonstrate enhanced cytotoxicity against mesothelin-positive cancer cells compared to unmodified NK-92 or control NK cells. Preclinical studies show promising in vitro and in vivo antitumor activity with limited side effects, suggesting potential for targeted immunotherapy of mesothelin-expressing malignancies[1].
