meso3 CAR-T is an experimental chimeric antigen receptor (CAR) T-cell therapy designed to target the membrane-proximal region (Region III) of mesothelin (MSLN). It is engineered using a modified, non-viral piggyBac transposon system. Preclinical research indicates that targeting the membrane-proximal region of MSLN (Region III) results in superior antitumor activity compared to targeting the distal region (Region I, often referred to as meso1). meso3 CAR-T cells demonstrate enhanced activation markers, such as CD107α, and increased production of pro-inflammatory cytokines including interleukin-2 (IL-2), TNF-α, and IFN-γ. In vivo studies using mouse models of gastric and ovarian cancer have shown that meso3 CAR-T cells can effectively inhibit tumor growth and mediate strong antitumor responses against MSLN-positive solid tumors.
