meso3-CD40 CAR-T is an experimental chimeric antigen receptor (CAR) T-cell therapy developed by researchers at Sun Yat-sen University for the treatment of mesothelin-expressing solid tumors, particularly ovarian cancer. The therapy utilizes the non-viral piggyBac transposon system to engineer T cells that target region III of mesothelin (MSLN), a membrane-proximal epitope. Uniquely, these CAR-T cells are also engineered to secrete anti-CD40 agonist antibodies, which are intended to activate the CD40 pathway and enhance the antitumor immune response within the suppressive tumor microenvironment. The CAR construct includes an anti-MSLN single-chain variable fragment (scFv), a CD28 transmembrane and co-stimulatory domain, and a CD3ζ signaling domain. Preclinical data indicate that meso3-CD40 CAR-T cells demonstrate superior cytokine production (including IFN-γ), increased central memory T-cell populations, and enhanced cytotoxic activity compared to standard mesothelin-targeted CAR-T cells.
