mesoCD19 CAR-TEAM is an experimental chimeric antigen receptor (CAR) T-cell construct designed as a research tool and control in the development of the CAR-TEAM (CAR-T cells secreting a T-cell-engaging molecule) platform. It consists of T cells engineered to express a CAR targeting mesothelin (meso) and to simultaneously secrete a T-cell-engaging molecule (TEAM) that targets CD19 and engages T cells via the CD3 epsilon subunit. In the context of solid tumor research, such as pancreatic ductal adenocarcinoma (PDAC) where CD19 is typically absent, this construct serves as a negative control to evaluate the specific contribution of the TEAM component when compared to therapeutic constructs targeting tumor-associated antigens like FAP. The TEAM technology is intended to overcome the immunosuppressive tumor microenvironment by engaging bystander T cells and targeting stromal components or heterogeneous tumor cells. The construct was developed primarily by researchers at Massachusetts General Hospital (Maus Lab).
