Overview

This combination cell therapy consists of autologous T-cells engineered to express chimeric antigen receptors (CARs) targeting two distinct tumor-associated antigens: Mesothelin (MSLN) and Claudin 18.2 (CLDN18.2). Developed by Essen Biotech, the treatment involves the sequential infusion of two separate CAR-T cell products following a lymphodepleting chemotherapy regimen of fludarabine and cyclophosphamide. Mesothelin is a cell surface glycoprotein overexpressed in various solid tumors, including pancreatic cancer, while Claudin 18.2 is a tight junction protein frequently expressed in gastric and pancreatic adenocarcinomas. By targeting both antigens, the therapy aims to address tumor heterogeneity and enhance therapeutic efficacy in patients with unresectable locally advanced or metastatic pancreatic ductal adenocarcinoma (PDAC).
