Mesothelin-specific chimeric antigen receptor (CAR) T cells are a form of adoptive cellular immunotherapy in which a patient's own T cells are genetically engineered to express a synthetic receptor that specifically recognizes mesothelin, a tumor-associated antigen highly expressed on the surface of various solid tumors such as malignant pleural mesothelioma, pancreatic cancer, ovarian cancer, and some lung and breast cancers. Upon reinfusion into the patient, these modified T cells can recognize and kill mesothelin-expressing tumor cells. The therapy is being investigated primarily for solid tumors with high mesothelin expression. Early-phase clinical trials have demonstrated safety and feasibility; efficacy remains under investigation. Some studies combine this therapy with immune checkpoint inhibitors like pembrolizumab to enhance antitumor activity[1][3][5].
