Overview

Mesothelin-targeted autologous CAR-T cells are an investigational cellular immunotherapy consisting of autologous T cells genetically engineered, typically via lentiviral transduction, to express a chimeric antigen receptor (CAR) targeting mesothelin (MSLN). Mesothelin is a cell-surface glycoprotein that is highly overexpressed in various malignant solid tumors, including mesothelioma, pancreatic cancer, ovarian cancer, and lung adenocarcinoma, while showing limited expression in normal tissues. Upon infusion, these CAR-T cells specifically bind to mesothelin-expressing tumor cells, leading to T-cell activation, proliferation, and targeted cytotoxicity. This therapy is being evaluated in clinical trials, such as those conducted by the Shanghai Jiao Tong University Affiliated Sixth People's Hospital, for the treatment of advanced mesothelin-positive malignant solid tumors.
