Mesothelin-targeted CAR-NK cells are an investigational adoptive cell therapy consisting of natural killer (NK) cells engineered to express a chimeric antigen receptor (CAR) specific for mesothelin (MSLN). Mesothelin is a cell surface glycoprotein that is highly expressed in several solid tumors, including epithelial ovarian cancer, mesothelioma, and pancreatic cancer, while maintaining limited expression in normal tissues. By targeting MSLN, these CAR-NK cells are designed to selectively bind to and eliminate tumor cells. NK cells offer potential advantages over CAR-T cells, such as a reduced risk of cytokine release syndrome (CRS) and graft-versus-host disease (GvHD), as well as innate anti-tumor mechanisms. Guojian Chengnuo Biotechnology is currently evaluating this therapy via intraperitoneal injection for the treatment of platinum-resistant epithelial ovarian cancer.
