Overview

messenger RNA encoding human cytomegalovirus glycoprotein L is a specific messenger RNA (mRNA) component of Moderna's cytomegalovirus (CMV) vaccine candidate, mRNA-1647. It encodes the glycoprotein L (gL) of the human cytomegalovirus. Glycoprotein L is a critical component of the viral entry machinery, forming a heterodimer with glycoprotein H (gH) and, in some cases, a pentameric complex with gH and the UL128/130/131A proteins. When delivered via lipid nanoparticles, this mRNA is translated by host cells to produce the gL protein, which then acts as an antigen to elicit a protective immune response, including the production of neutralizing antibodies. This approach is designed to prevent CMV infection, particularly in seronegative women of childbearing age to prevent congenital CMV. The vaccine candidate mRNA-1647, which includes this mRNA along with five others, is currently in Phase 3 clinical development.
