The messenger RNA TEM1 cancer vaccine is an experimental immunotherapy designed to target the Tumor Endothelial Marker 1 (TEM1), also known as CD248. TEM1 is a cell surface glycoprotein that is highly expressed on tumor-associated pericytes and fibroblasts within the tumor microenvironment, as well as on certain malignant cells such as sarcomas, while remaining largely absent in normal adult tissues. The vaccine consists of mRNA encoding the TEM1 protein, typically encapsulated in lipid nanoparticles (LNPs) for delivery. Upon administration, host cells translate the mRNA into TEM1 protein, which serves as an antigen to stimulate a robust T-cell-mediated immune response. This response is intended to selectively destroy TEM1-expressing cells, thereby disrupting the tumor's vascular support and stroma, inhibiting tumor growth, and potentially enhancing the infiltration of other therapeutic agents. The vaccine has been primarily developed and investigated by researchers at the University of Pennsylvania and the Wistar Institute.
