MET-233 (also referred to as MET-233i) is an ultra-long acting amylin analog developed by Metsera for the treatment of obesity. It is a synthetic peptide engineered to mimic amylin, a hormone co-secreted with insulin that regulates appetite and increases satiety. The drug leverages Metsera’s proprietary HALO (Half-Life Optimization by Lipid Optimization) technology, which extends its half-life to 19 days—enabling potential once-monthly subcutaneous dosing. In Phase 1 clinical trials involving overweight or obese adults without type 2 diabetes, MET-233 demonstrated up to 8.4% mean placebo-subtracted weight loss at Day 36 after five weekly doses of 1.2 mg, with some individuals achieving up to a 10.2% reduction in body weight. The safety profile was favorable; gastrointestinal side effects were mild and dose-dependent, resolving within the first week of dosing and no severe adverse events were reported[3][4][5][6][7][8].
