MET-CAR.4-1BBζ is a chimeric antigen receptor T cell (CAR-T) therapy engineered to target cancer cells that overexpress the MET (c-MET) receptor tyrosine kinase. The CAR constructs are based on the single-chain variable fragment (scFv) derived from MetMab (an anti-MET antibody), fused to a 4-1BB (CD137) co-stimulatory domain and a CD3ζ (zeta chain) signaling domain. Upon binding MET on the surface of tumor cells, the CAR-T cells become activated, resulting in targeted cell lysis and cytokine secretion. MET-CAR.4-1BBζ was developed for hepatocellular carcinoma (HCC) and functions independently of MET pathway activation, allowing it to target MET-overexpressing tumors regardless of the activation or phosphorylation status of MET. Preclinical studies support its activity in vitro and in mouse xenograft models, where it demonstrates anti-tumor efficacy, T cell activation, and cytokine production[1][3].
