Opioid Growth Factor (OGF), chemically known as met-enkephalin ([Met5]-enkephalin), is an endogenous pentapeptide biologic being investigated by the University of Missouri-Columbia for the treatment of unresectable hepatocellular carcinoma (HCC) in patients with cirrhosis. OGF functions as a negative growth regulator by binding to the Opioid Growth Factor Receptor (OGFr), which forms an endogenous growth-regulating pathway. This interaction exerts a dose-dependent, reversible, and receptor-mediated inhibitory action on cancer cell proliferation, specifically through the induction of the cyclin-dependent kinase inhibitory pathway. The drug has been evaluated in Phase I clinical trials (such as NCT00706576) where it was administered via intravenous infusion to assess safety, toxicity, and pharmacokinetics in patients with advanced liver disease and underlying cirrhosis.
