Overview

Metadherin siRNA is a small interfering RNA (siRNA) designed to silence the expression of metadherin (MTDH), also known as astrocyte elevated gene-1 (AEG-1). MTDH is an oncogene that is frequently overexpressed in various malignancies, including head and neck squamous cell carcinoma (HNSCC) and nasopharyngeal carcinoma (NPC), where it promotes tumor progression, epithelial-mesenchymal transition (EMT), and metastasis. The siRNA works through the RNA interference pathway to degrade MTDH mRNA, thereby reducing protein levels and inhibiting downstream signaling cascades such as the PI3K/Akt pathway. Preclinical studies have demonstrated that knockdown of MTDH using siRNA significantly reduces cancer cell viability, migration, invasion, and in vivo tumor growth, suggesting that targeting this pathway could be a novel therapeutic strategy for improving outcomes in patients with head and neck cancers.
