Metadoxine is a small molecule drug used primarily for the treatment of acute and chronic alcohol intoxication, alcoholic fatty liver disease, non-alcoholic fatty liver disease, and cholestatic liver diseases. It acts as a hepatoprotective agent by enhancing the elimination of alcohol from blood and tissues, accelerating ethanol metabolism through increased activity of aldehyde dehydrogenase (ALDH), and protecting hepatocytes from oxidative stress. Metadoxine also modulates gamma-aminobutyric acid (GABA) transmission by inhibiting GABA transaminase activity and increasing inhibitory GABAergic synaptic transmission. Additionally, it prevents glutathione depletion, lipid peroxidation damage, collagen deposition, TNF-α induction in hepatocytes exposed to alcohol or acetaldehyde, and may prevent progression to cirrhosis in chronic exposure models[1][2][3][5][6][8].
