Overview
MG01CI is an extended-release formulation of metadoxine (pyridoxine-L-pyroglutamate), a non-stimulant small molecule originally developed by Alcobra Pharmaceuticals for the treatment of attention-deficit hyperactivity disorder (ADHD) and Fragile X syndrome. Metadoxine acts as a selective antagonist at the 5-HT2B receptor and modulates GABAergic transmission, offering a mechanism distinct from traditional dopaminergic stimulants. While metadoxine has been used for decades in certain regions for acute alcohol intoxication, MG01CI was designed to provide sustained plasma levels suitable for cognitive disorders. Development for ADHD was discontinued following the failure of the Phase III MEASURE trial to meet its primary endpoint in 2017, which also led Teva Pharmaceutical Industries to decline its option to acquire Alcobra.
