Overview

Metelimumab is a human IgG4 monoclonal antibody that selectively neutralizes transforming growth factor beta 1 (TGF-β1), developed as a TGF-β1-specific antagonist using phage display technology at Cambridge Antibody Technology and later partnered with Genzyme.[1][9][11] By binding TGF-β1 and preventing receptor engagement, it was intended to reduce pathological fibrosis and immune dysregulation, and was clinically investigated primarily in diffuse cutaneous systemic sclerosis (scleroderma), where excess TGF-β1 signaling drives abnormal extracellular matrix deposition and tissue fibrosis, but its clinical development was terminated after phase I/II trials showed acceptable safety but insufficient efficacy and a higher rate of adverse events versus placebo.[1][2][8][9][11]
