Overview

Metformin hydrochloride + glibenclamide is a fixed-dose combination pharmaceutical product used for the management of type 2 diabetes mellitus. It combines two antihyperglycemic agents with distinct but complementary mechanisms of action: metformin hydrochloride, a biguanide, and glibenclamide (also known as glyburide), a second-generation sulfonylurea. Metformin primarily works by decreasing hepatic glucose production, reducing intestinal absorption of glucose, and improving insulin sensitivity by increasing peripheral glucose uptake and utilization. Glibenclamide acts by stimulating the release of insulin from functioning pancreatic beta cells through the inhibition of ATP-sensitive potassium channels. This combination is typically indicated as an adjunct to diet and exercise to improve glycemic control in adults with type 2 diabetes when treatment with either agent alone is insufficient. The reference branded product, Glucovance, was originally developed by Bristol Myers Squibb.
