metformin + henagliflozin + retagliptin

This refers to the sequential treatment arm of a Phase 4 clinical trial (NCT05614401) sponsored by Peking University First Hospital, designed to evaluate the efficacy and safety of initial triple combination therapy versus a traditional stepwise approach in patients with newly diagnosed type 2 diabetes mellitus. The regimen involves the sequential administration of three glucose-lowering agents: metformin (a biguanide), henagliflozin proline (an SGLT2 inhibitor), and retagliptin (a DPP-4 inhibitor). Patients in this group begin with metformin monotherapy; if glycemic targets (fasting blood glucose >7 mmol/L or postprandial blood glucose >10 mmol/L) are not achieved, henagliflozin is added, followed by retagliptin if control remains inadequate. Henagliflozin and retagliptin are small molecule inhibitors developed by Jiangsu Hengrui Pharmaceuticals.
