Overview
This combination therapy consists of metformin, orlistat, and pioglitazone, utilized as a standard-of-care control arm in clinical research conducted by the Affiliated Hospital of Nantong University. It is specifically evaluated for the treatment of patients with Polycystic Ovary Syndrome (PCOS) and comorbid Type 2 Diabetes Mellitus (T2DM). Metformin serves as an insulin sensitizer by reducing hepatic glucose production and improving peripheral glucose uptake; pioglitazone, a thiazolidinedione, further enhances insulin sensitivity via the activation of the peroxisome proliferator-activated receptor gamma (PPAR-gamma); and orlistat, a lipase inhibitor, is included for obese patients to manage weight by preventing the absorption of dietary fats. This multi-modal pharmacological approach addresses the complex metabolic and endocrine dysfunctions, such as insulin resistance and obesity, that characterize the clinical presentation of PCOS and T2DM.
