Overview

Metformin + rapamycin is an experimental **combination therapy** comprising two small molecule drugs: metformin, a well-established AMP-activated protein kinase (AMPK) activator and first-line antidiabetic medication, and rapamycin, a mammalian target of rapamycin (mTOR) inhibitor with immunosuppressive and antineoplastic properties. This combination is being explored for its potential to synergistically inhibit mTOR signaling, reduce PI3K/AKT pathway activity, and decrease cell proliferation. Together, the drugs aim to enhance anti-cancer, anti-aging, metabolic, and potentially anti-viral effects compared to either agent alone. Preclinical studies show promising synergy in oncology, while early-phase studies and patents propose their use in diseases related to aberrant mTOR signaling, including cancer, metabolic syndrome, neurodegenerative, inflammatory, and autoimmune disorders.
