Metformin eicosapentaenoate (TP-101) is a novel small molecule salt comprising metformin, a biguanide antihyperglycemic, and eicosapentaenoic acid (EPA), an omega-3 fatty acid. Developed by Thetis Pharmaceuticals, it is intended for the treatment of type 2 diabetes mellitus and hypertriglyceridemia. The drug utilizes a proprietary HEAL (High Efficiency Amino Lipid) technology to ionically bond metformin and EPA into a single chemical entity. Metformin primarily acts by inhibiting hepatic glucose production and increasing peripheral glucose uptake via AMPK activation, while EPA reduces serum triglycerides by inhibiting hepatic VLDL synthesis and enhancing fatty acid oxidation. TP-101 is designed to provide the therapeutic benefits of both agents with improved stability and potentially enhanced bioavailability compared to co-administration of the individual components.
