Drug intelligence / Profile preview
Metformin gum is an experimental oral formulation of the biguanide antihyperglycemic agent metformin, developed by researchers at the Hadassah Medical Organization. This novel delivery system is designed as a medicated chewing gum to provide an alternative to traditional oral tablets, potentially improving patient compliance or altering the pharmacokinetic profile of the drug. Metformin's primary therapeutic effect is achieved through the activation of AMP-activated protein kinase (AMPK) and the inhibition of mitochondrial complex I, which collectively result in decreased hepatic glucose production (gluconeogenesis) and enhanced insulin sensitivity in peripheral tissues such as skeletal muscle. The formulation was evaluated in a Phase 1 clinical trial to compare its bioavailability and pharmacokinetics against standard immediate-release metformin tablets in healthy volunteers.
