Metformin METOral is an oral nanoparticle formulation of the biguanide antihyperglycemic agent metformin, developed by Nanomerics using its proprietary Molecular Envelope Technology (MET). The MET platform utilizes amphiphilic polymers, specifically N-palmitoyl-N-monomethyl-N,N-dimethyl-N,N,N-trimethyl-6-O-glycolchitosan (GCPQ), to encapsulate small molecules into nanoparticles. This delivery system is designed to protect the drug from the gastrointestinal environment and facilitate its transport across the intestinal mucus and epithelial barriers, thereby enhancing oral bioavailability and potentially reducing gastrointestinal side effects. Metformin itself acts primarily by inhibiting mitochondrial complex I and indirectly activating AMP-activated protein kinase (AMPK), which leads to a reduction in hepatic gluconeogenesis and an improvement in peripheral insulin sensitivity. The metformin METOral program is currently in preclinical development for multiple potential indications.
