Overview

Methacycline (also known as metacycline) is a broad-spectrum semisynthetic antibiotic in the tetracycline class. It is structurally related to oxytetracycline and acts as a protein synthesis inhibitor by binding reversibly to the 16S rRNA of the 30S ribosomal subunit in bacteria, thereby preventing aminoacyl-tRNA from binding to the A site of the ribosome and inhibiting translation. Methacycline exhibits activity against a wide range of Gram-positive and Gram-negative bacteria, as well as some protozoa. It has been used for various bacterial infections including respiratory tract infections and urinary tract infections. Methacycline is also notable for its slower excretion compared to other tetracyclines, resulting in prolonged effective blood levels. Additionally, it serves as an industrial precursor for doxycycline hyclate synthesis[1][2][3][4][6].
