Drug intelligence / Profile preview
Methionine gamma-lyase + daidzein is an investigational bioconjugate consisting of the enzyme methionine gamma-lyase (MGL) covalently linked to daidzein, a soy isoflavone. This drug is typically used as part of a pharmacological pair in directed enzyme prodrug therapy, where MGL-Dz catalyzes the in situ conversion of a sulfur-containing substrate (such as S-propyl-L-cysteine sulfoxide, propiin) into cytotoxic thiosulfinates within tumors. The mechanism leverages the methionine-depleting and thiosulfinate-generating properties of MGL to induce selective cytotoxicity in tumor cells, while the daidzein conjugation enhances tumor targeting or pharmacokinetics. Preclinical studies demonstrate antitumor effects in several solid tumor models, including colon, pancreatic, and prostate cancers, with significant inhibition of tumor growth in murine xenograft models[1][3].
