Methionine sulfoximine is a synthetic, non-proteinogenic amino acid and the sulfoximine derivative of methionine. It acts as an irreversible inhibitor of glutamine synthetase (EC 6.3.1.2), leading to decreased synthesis of glutamine from glutamate and ammonia[3][4][5]. This inhibition disrupts the astrocyte-neuron glutamate shuttle, reducing neuronal glutamate release and potentially preventing excitotoxicity in neural tissue[3][7]. Methionine sulfoximine has convulsant effects in some species but can be neuroprotective at sub-convulsive doses in rodent models of hyperammonemia, acute liver disease, amyotrophic lateral sclerosis (ALS), and cerebral ischemia[4][7]. It is primarily used as a research tool rather than a therapeutic agent.
