Methotrexate (nanoparticle reformulation) is a preclinical-stage antineoplastic agent being developed by Prosemble. It consists of a nanoparticle-based delivery system for methotrexate, a small molecule inhibitor of dihydrofolate reductase (DHFR). By inhibiting DHFR, the drug prevents the conversion of dihydrofolate to tetrahydrofolate, thereby disrupting DNA synthesis and cell proliferation. The nanoparticle formulation is designed to improve the pharmacokinetic profile and tumor-specific delivery of methotrexate, potentially enhancing efficacy in leukaemia, lymphoma, and solid tumours while minimizing systemic off-target toxicities.
