Overview

The MMOPP regimen is a combination chemotherapy protocol evaluated by the M.D. Anderson Cancer Center, specifically in a Phase II clinical trial (NCT00002463). This regimen was designed as a primary therapy for infants and young children with primitive neuroectodermal tumors (PNET) or high-grade astrocytomas to delay or avoid the need for cranial irradiation. It integrates high-dose methotrexate (HMTX) with the standard MOPP protocol, which consists of mechlorethamine, vincristine, procarbazine, and prednisone. Methotrexate acts as an antimetabolite by inhibiting dihydrofolate reductase, thereby disrupting DNA synthesis. Mechlorethamine and procarbazine serve as DNA alkylating agents, vincristine acts as a microtubule inhibitor to arrest mitosis, and prednisone is a corticosteroid that provides lympholytic effects and reduces cerebral edema.
