Methotrexate + sulfasalazine is a combination therapy of two disease-modifying antirheumatic drugs (DMARDs) used primarily in the treatment of rheumatoid arthritis. This combination has been shown to be more effective than either drug alone, particularly in patients who have had a suboptimal response to sulfasalazine monotherapy. The combination works through complementary anti-inflammatory mechanisms, with methotrexate inhibiting dihydrofolate reductase and sulfasalazine having anti-inflammatory effects through mechanisms that may include folate depletion.
