Methotrexate + trastuzumab is a combination therapy used for HER2-positive breast cancer. Trastuzumab is a monoclonal antibody that targets the HER2 receptor, which is overexpressed in approximately 20-30% of breast cancers. Methotrexate is an antifolate drug that inhibits purine de novo synthesis, impeding DNA synthesis and repair. The combination has shown efficacy in metastatic breast cancer patients, including those previously treated with trastuzumab. Methotrexate can enhance the cytotoxicity of trastuzumab-based therapies by acting as a radiosensitizer.
