Methotrexate + vobarilizumab is a combination therapy investigated for the treatment of moderate-to-severe rheumatoid arthritis (RA). Methotrexate is a folate analog antimetabolite and immunosuppressant that inhibits enzymes involved in nucleotide synthesis, leading to anti-inflammatory effects primarily through increased adenosine release and inhibition of transmethylation reactions. Vobarilizumab is a humanized Nanobody® targeting the interleukin-6 receptor (IL-6R), blocking IL-6 signaling and thereby reducing inflammation. The combination has been studied in phase 2b clinical trials for patients with active RA who had an inadequate response to methotrexate alone, showing positive impacts on disease activity and safety profile[1][4][5][10].
