Drug intelligence / Profile preview
Methotrexate + zinc is a combination therapy consisting of the antimetabolite and immunosuppressant methotrexate co-administered with zinc (often in the form of zinc sulfate, zinc gluconate, zinc aspartate, or zinc citrate). Methotrexate acts as a dihydrofolate reductase (DHFR) inhibitor, suppressing DNA synthesis and cell proliferation, which is widely used to treat autoimmune diseases like rheumatoid arthritis and psoriasis, as well as various cancers. Zinc is an essential micronutrient and antioxidant that regulates immune function, bone remodeling, and inflammatory pathways. Co-administration of zinc with methotrexate is primarily investigated to enhance therapeutic efficacy in rheumatoid arthritis and psoriasis, and to mitigate methotrexate-induced toxicities, such as gastrointestinal toxicity, mucositis, hepatotoxicity, and neurotoxicity, by reducing oxidative stress and inflammation.
