Overview

MTX-ATMPs are a novel therapeutic modality consisting of the chemotherapeutic agent methotrexate (MTX) packaged within autologous tumor-derived microparticles (ATMPs). Developed by researchers at Union Hospital, Tongji Medical College, this approach utilizes the microparticles as a delivery vehicle that tumor cells naturally internalize via endocytosis. This mechanism allows the drug to bypass common resistance pathways, such as drug efflux pumps, and specifically target tumor-repopulating cells (TRCs). Once inside the malignant cell, the methotrexate is released to inhibit dihydrofolate reductase, leading to the disruption of DNA synthesis and subsequent cell death. The therapy is primarily investigated for the treatment of malignant pleural effusion (MPE), where it is administered intrapleurally to clear malignant cells and potentially stimulate a localized anti-tumor immune response.
