Methotrexate-loaded tumor cell-derived microparticles (MPs-MTX or TMPs-MTX) is an innovative drug delivery system and immunotherapeutic agent developed by Peking Union Medical College for the treatment of malignant pleural effusion (MPE). This therapy utilizes tumor-derived extracellular vesicles (microparticles) as natural carriers to encapsulate the chemotherapeutic agent methotrexate. Unlike traditional systemic chemotherapy, MPs-MTX is administered via intrapleural infusion and acts by mobilizing and activating neutrophils within the pleural space. These activated neutrophils then exert potent anti-tumor effects, making the drug an effective treatment for MPE secondary to advanced non-squamous non-small cell lung cancer or breast cancer. The microparticles serve a dual role as both a delivery vehicle for the cytotoxic payload and a biological stimulus for the local immune environment.
