Overview

Methyclothiazide is a small molecule, thiazide diuretic and antihypertensive medication. It works by inhibiting the sodium-chloride symporter (NCC) in the distal convoluted tubules of the kidneys, leading to increased excretion of sodium, chloride, and water, thereby reducing blood volume and blood pressure. It also possesses mild vasodilatory properties. Primarily indicated for the treatment of hypertension and edema associated with conditions such as congestive heart failure, hepatic cirrhosis, kidney disease, and corticosteroid/estrogen therapy, it has also been used for diabetes insipidus, certain electrolyte disturbances, and to prevent kidney stones. Methyclothiazide was first approved in the United States in 1960 and was developed and marketed by AbbVie, Inc., though it is currently discontinued in the U.S.
