Overview

Methylphenidate + ondansetron (also known as OND-PR002 or MPh-IR + Ond-PR2) is an investigational fixed-dose combination of the central nervous system stimulant methylphenidate and the selective 5-HT3 serotonin receptor antagonist ondansetron. Developed by Duke University and its spinout Generys Biopharmaceuticals, the combination is designed to treat substance use disorders (such as cocaine and methamphetamine use disorders) and trauma- and stressor-related disorders. The therapeutic strategy relies on 'pharmacologically-mediated reactivation and reconsolidation blockade' of dysfunctional neural memories underlying addiction. The formulation combines immediate-release methylphenidate (acting as a dopamine and norepinephrine reuptake inhibitor to reactivate the drug memory) with a delayed pulsatile-release formulation of ondansetron (to block memory reconsolidation). It has been evaluated in Phase IIa clinical trials.
